Research achievement of Professor Li Lei was published in CELL Research

Recently, SCI paper of "Single-cell exome sequencing identifies mutations in KCP, LOC440040, and LOC440563 as drivers in renal cell carcinoma stem cells" with Li Lei, Professor of Urology Department of our hospital, as the corresponding author and our hospital as the first institution was published in Cell Research issued on December 16. The impact factor of this peiodical reached up to 14.8. The key point of the article was to identify KCP, LOC440040 and LOC440563 mutations as drivers of renal cell carcinoma stem cells by single-cell exome sequencing.

In the article, single-cell exome sequencing was first used to evaluate the somatic mutations of tumors, which was of far-reaching significance for guiding clinical practice in the future.

It is found in this research that CD133+ RCC cells are of the feature of CSC and may derive from cancer cells instead of normal renal cells. KCP, LOC440040 and LOC440563 mutations are driving genes of new renal carcinoma stem cells. LOC440563 encodes a RNA binding protein, which belongs to a heterogeneous nuclear ribonucleoprotein (hnRNPs) and affects processing, metabolism and transportation of precursor mRNAs. LOC440563 is of significant mutations in colon cancer. However, these gene mutations are different from those identified in RCC carcinoma stem cells. Taking the contributions of hnRNP family to repair of DNA damage into consideration, it is possible that the LOC440563 mutation drives pluripotency of CSC by damaging the repair process of DNA.
